Description :
CD28H (CD28 homolog), also known as TMIGD2 (transmembrane and immunoglobulin domain-containing protein 2) and IGPR1 (immunoglobulin and proline-rich receptor 1), is a single-pass type I transmembrane glycoprotein. CD28H contains 1 Ig (immunoglobulin)-like domain in the extracellular region and 1 cytoplasmic proline-rich domain. CD28H is widely expressed, mainly by epithelial and endothelial cells. It is also expressed in lymphoid organs, with highest levels in thymus, spleen, peripheral blood lymphocytes and liver. In the thymus, it is expressed in CD4 and CD8 single- and double-positive cells, but not in immature CD4 and CD8 double-negative cells. In peripheral blood mononuclear cells, CD28H is highly expressed on natural killer (NK) cells and CD3+ T-cells but not on B-cells. CD28H may form homophilic interactions that could regulate cell-cell interaction, cell migration, and angiogenesis. The B7 family member B7-H7 (B7 homolog 7), also know n as HHLA2 (human endogenous retrovirus-H long terminal repeat-associating protein 2), was identified as a specific ligand for CD28H. Through interaction with B7-H7, CD28H may co-stimulate T-cells in the context of TCR-mediated activation. The B7-H7/CD28H interaction selectively co-stimulates human T-cell growth and cytokine production via an AKT-dependent signalling cascade.
