Description :
The leukocyte adhesion molecule CD226, also known as DNAM1 (DNAX accessory molecule1) and PTA1 (platelet and T cell activation antigen 1), is a single-pass type I transmembrane glycoprotein belonging to the immunoglobulin (Ig) superfamily (IgSF). CD226 contains a 236 amino acid (aa) extracellular domain (ECD) with 2 Iglike C2set (IgC2) domains and a 61 aa cytoplasmic region that contains motifs for binding PDZ domains and band 4.1 family proteins. CD226 is identified as an intercellular adhesion molecule, which is constitutively expressed on the majority of CD4+ and CD8+ T lymphocytes, natural killer (NK) cells, monocytes/macrophages, and a subset of B lymphocytes. CD226 is expressed on multiple lymphoid and myeloid cells and interacts with the poliovirus receptor CD155/PVR and its family member CD112/Nectin2. Ligation of CD226 promotes the activation of NK cells, CD8+ T cells, and mast cells, dendritic cell maturation, megakaryocyte and activated platelet adhesion to vascular endothelial cells, and monocyte extravasation; but inhibits the forrmation of osteoclasts. CD226 is critical for NK cell- and CD8+ T cell-mediated cytotoxicity. It also plays an important role in immune surveillance of tumor development and is involved in triggering naive T cell differentiation and proliferation. Plateletendothelium interactions mediated by CD226 may contribute to the metastasis of tumor cells to the lung.
