Description :
LTβR (Lymphotoxin beta receptor), also known as TNFR3 or TNF Rrp, is a single-pass type I transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF3). LTβR contains4 cysteine-rich repeats within the extracellular domain (ECD) characteristic of the TNFRSF. LTβR ligands include homotrimeric of LIGHT/TNFSF14 and heterotrimeric lymphotoxin-α1/β2. LTβR is expressed on the surface of most cell types, including cells of epithelial and myeloid lineages, but not on T and B lymphocytes. LTβR is expressed on mesenchymal stromal organizing cells that give rise to stroma of primary (thymus), secondary (tonsils, lymph nodes and Peyers patches) and tertiary (ectopic inflammatory) lymphoid structures. Secondary immune tissues are absent in LTβR-deficient mice. Depending on the cell type, activation of LTβR has been shown to induce canonical or alternative NF-κB pathways. Soluble LTβR can be formed by proteolytic cleavage of the ECD, and act as an antagonist to inhibit autoimmunity. LTβR regulates cell growth and can initiate inflammation-related carcinogenesis. It is expressed in hepatocytes, is up-regulated in regeneration, hepatitis and hepatocellular carcinoma, and influences lipid metabolism and atherosclerosis.

The recombinant human LTβR-Fc fusion is expressed as a 420 amino acid protein consisting of Gln31 – Thr234 region of LTβR (UniProt accession #P36941) and a C-terminal Fc from human IgG1, which exists as a dimer/tetramer/oligomer under non-reducing conditions (see the gel image inserted above).
