Description :
DcR1 ( decoy receptor 1), also known as CD263, TRAILR3, LIT and TRID, is a glycosylphosphatidylinositol (GPI)linked membrane protein of the TNF receptor superfamily (TNFRSF10C). DcR1 is highly expressed in peripheral blood lymphocytes, spleen, skeletal muscle, placenta, lung and heart. DcR1 is one of 5 receptors for TRAIL (tumor necrosis factor-related apoptosis inducing ligand), also known as Apo2L (Apo-2 ligand) and TNFSF10. TRAIL preferentially induces apoptosis in tumor cell lines, but not in normal cells. In addition to DcR2, DcR3 and OPG, DcR1 is another TNFRSF member that modulates cytotoxic ligands-induced apoptosis. DcR1 binds TRAIL with high affinity. However DcR1 lacks a cytoplasmic death domain and hence is not capable of inducing apoptosis. DcR1 may protect cells against TRAIL mediated apoptosis by competing with DR4/TRAILR1 and DR5/TRAILR2 for binding to the ligand. Expression of DcR1 has been shown to protect cells bearing DR4 and/or DR5 from TRAILinduced apoptosis. DcR1 is also a p53-regulated DNA damage-inducible gene. DcR1 is detected in many normal tissues but not in most cancer cell lines, which may explain the specific sensitivity of cancer cells to the apoptosis-inducing activity of TRAIL. The soluble DcR1-Fc fusion protein neutralizes the ability of TRAIL to induce apoptosis.

The recombinant human DcR1-Fc fusion is expressed as a 438 amino acid protein consisting of Ala26 – Pro235 region of DcR1 (UniProt accession #O14798) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted).
