Description :
CD40 s a single-pass, type I transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF5). CD40 is expressed on the surface of B cells, dendritic cells, macrophages, monocytes and platelets, as well as endothelial and epithelial cells. CD40 plays an essential role in mediating a broad range of immune and inflammatory responses including immunoglobulin class switching, B cell development, and germinal center formation. CD40 contains 4 cysteine-rich repeats in the extracellular region. Interaction of CD40 with its ligand, CD40 Ligand (CD40L, also known as CD154), leads to the aggregation of CD40 molecules resulting in the initiation of bidirectional intracellular signaling in both CD40 and CD40L expressing cells. CD40 ligation by CD40L promotes B cell activation and T celldependent humoral responses. Mutations in CD40 are associated with hyper-IgM immunodeficiency type 3 (HIGM3), an autosomal recessive disorder characterized by an inability of B cells to undergo isotype switching or to mount an antibody-specific immune response. CD40 is also implicated in the pathology of multiple cardiovascular diseases, such as atherosclerosis, atherothrombosis, and restenosis. CD40/CD40L interaction may be required for amyloid-beta-induced microglial activation, an early event in Alzheimer disease pathogenesis. In addition CD40 plays important roles in both hematopoietic and epithelial cancers. CD40 has been a target for tumor immunotherapy.

The recombinant human CD40-Fc fusion is expressed as a 401 amino acid protein consisting of Glu21 – Arg193 region of CD40 (UniProt accession #P25942) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted above).
