Description :
DcR3 (death decoy receptor), also known as decoy receptor for Fas ligand, TR6 and M68, is a member of the TNF receptor superfamily (TNFRSF6B). Like osteoprotegerin (OPG), DcR3 lacks a transmembrane sequence and is a secreted protein that contains 4 TNFR cysteine-rich repeats. Expression of DcR3 has been reported in a wide array of normal tissues as well as primary tumors and tumor cell lines. DcR3 is detected in fetal lung, brain and liver and in adult stomach, spinal cord, lymph node, trachea, spleen, colon and lung. DcR3 is highly expressed in primary tumors from colon, stomach, rectum, and esophagus as well as in SW480 colon cancer cell line. DcR3 was shown to bind with Fas ligand (FasL) and LIGHT and TNF- like molecule 1A (TL1A). The principal function of DcR3 is the inhibition of FasL­ and LIGHT-­induced apoptosis. In addition to DcR1, DcR2 and OPG, DcR3 is another TNFRSF member that modulates cytotoxic ligands-induced apoptosis. Overexpression of DcR3 may be a mechanism by which certain tumors escape immune­cytotoxic attack. DcR3 may also modulate CNS- autoimmunity by interfering with Th17 responses via the blockade of Fas-FasL interaction.

The recombinant human DcR3-Fc fusion is expressed as a 499 amino acid protein consisting of Val30 – His300 region of DcR3 (UniProt accession #O95407) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted above).
