Description :
T-cell surface glycoprotein CD3 epsilon chain, also known as CD3e, T3e, TCRe and IMD18, is a single-pass type I transmembrane protein. CD3e contains 1 Ig-like (immunoglobulin-like) domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3 gamma chain, also known as CD3g, T3g and IMD17, is a single-pass type I transmembrane protein. CD3g contains 1 Ig-like domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3e:CD3g is part of the T-cell receptor (TCR)/CD3 complex and is involved in T-cell development and signal transduction. The TCR/CD3 complex of T-lymphocytes consists of either a TCR alpha/beta or TCR gamma/delta heterodimer co-expressed at the cell surface with the invariant subunits of CD3 gamma, delta, epsilon, zeta, and eta. The TCR/CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways leading to the activation of T cells. CD3e plays an essential role in T-cell development. In CD3e-deficient mice, all stages of T cell development and activation that are TCR-dependent were impaired, but not eliminated. Mutations in the CD3e gene are associated with immunodeficiency 18 (IMD18), an autosomal recessive primary immunodeficiency characterized by onset in infancy or early childhood. Defects in the CD3g gene are a cause of immunodeficiency 17 (IMD17), an autosomal recessive primary immunodeficiency characterized by highly variable clinical severity.

Construct Details :
The recombinant human CD3e-CD3g heterodimer is co-expressed as 2 chains: (1) a 326-amino acid CD3e-Fc chain consisting of Gln22 – Val118 region of CD3e and a C-terminal Fc from human IgG1; (2) a 327-amino acid CD3g-Fc fusion consisting of Gln23 – Met103 region of CD3g and a C- terminal Fc from human IgG1. It exists as a dimer of heterodimer under non-reducing condition.
