Description :
HVEM (herpesvirus entry mediator), also referred to as CD270, TR2 and ATAR, is a single-pass, type I transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF14). HVEM is widely expressed, with the highest expression in lung, spleen and thymus. It is found in peripheral blood T cells, B cells, monocytes and in various tissues enriched in lymphoid cells. The extracellular domain (ECD) of HVEM contains three cysteine­rich repeats characteristic of TNF receptor superfamily members. The ECD of HVEM interacts directly with the herpes simplex virus (HSV) envelope glycoprotein D, contributing partly to HSV entry mechanism. The short cytoplasmic region of HVEM lacks a death domain, but contains a TRAF interaction motif for recruiting TRAFs and transducing signaling pathways that activate inflammatory and inhibitory T-cell immune responses. Two TNF superfamily ligands lymphotoxin-α (also known as TNF-β) and LIGHT (TNFSF14) are identified as cellular ligands for HVEM. Both ligands initiate the positive signaling activities. However, recent studies have also revealed that HVEM may be involved in the inhibitory pathway for T cells through serving as a receptor for BTLA (B and T lymphocyte attenuator) or CD272.

The recombinant human HVEM-Fc fusion is expressed as a 388 amino acid protein consisting of Leu39 – Ser199 region of HVEM (UniProt accession #Q92956) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted above).
